Bicara Therapeutics has announced a significant leadership transition as it prepares for a critical phase in the development of its lead drug candidate. The company, listed on the Nasdaq under the ticker BCAX, stated on 11 August that current CEO Claire Mazumdar will move into a Vice Chair and Strategic Adviser role at the start of next year. She will be succeeded by Ryan Cohlhepp, who currently serves as President and Chief Operating Officer.

The change in leadership comes at a pivotal moment for the firm, which recently presented three-year follow-up data for ficerafusp alfa, a treatment for HPV-negative head and neck cancer. Data shared at the May ASCO meeting indicated that at the 1,500 milligram weekly dose, approximately one in three patients remained alive at three years. Management noted this figure was roughly double the survival rate observed in retrospective analyses of standard-of-care pembrolizumab in the same patient population.

Bicara’s pivotal Phase III FORTIFI-HN01 study is on track for substantial enrolment by the end of 2026, with more than 200 sites currently active. An interim readout is expected in mid-2027, which could potentially open the door to accelerated approval in the United States. The company has also launched FORTIFI-FLEX, a study investigating an every-three-week maintenance dose, with data targeted for release around the time of any US approval decision.

The executive shake-up extends beyond the CEO position. Jenn Larson has taken over as Chief Financial Officer, succeeding Ivan Hyep, while Chief Development Officer Tanya Green will move into the Chief Operating Officer seat and Chief Corporate Affairs Officer Jenna Cohen will become Chief Business Officer, both effective in January. A new Chief Legal Officer and two additional board members are also joining the company, concentrating a substantial amount of organisational change in a single quarter.

Financially, the company reported that operating expenses rose in the second quarter of 2026 compared with the same period the previous year. This increase is driven by clinical operations, manufacturing costs, and headcount growth. Management expects expenses to continue climbing quarter over quarter for the remainder of the year as it builds out medical affairs and commercial teams, all before the drug has generated revenue.

Bicara currently holds approximately $497 million in cash, which management states is sufficient to fund operations through the first half of 2029. Institutional interest remains steady, with hedge fund ownership holding flat at 24 funds in the most recent quarter, while short interest sits at 20.4% of the float.
